Looking at the PP article on how to calibrate your TPS switch with an ohm meter.

When doing this, do I unplug the TPS from the harness or just connect up my ohm meter through the "back" of the harness plug?

I would think unplugged so as not to get signals through the harness. Alligator clips attached to the pins in the plug part on the TPS... If I remember correctly, you want to know when contact is made so it's a continuity check on the meter.

You unplug. Else it is hard to do the test because you dont have access to the terminals. It is easier than it sounds and pretty simple once you get going.
